Brain circuits for spatial navigation

In this webinar on spatial navigation circuits, three researchers—Ann Hermundstad, Ila Fiete, and Barbara Webb—discussed how diverse species solve navigation problems using specialized yet evolutionarily conserved brain structures. Hermundstad illustrated the fruit fly’s central complex, focusing on how hardwired circuit motifs (e.g., sinusoidal steering curves) enable rapid, flexible learning of goal-directed navigation. This framework combines internal heading representations with modifiable goal signals, leveraging activity-dependent plasticity to adapt to new environments. Fiete explored the mammalian head-direction system, demonstrating how population recordings reveal a one-dimensional ring attractor underlying continuous integration of angular velocity. She showed that key theoretical predictions—low-dimensional manifold structure, isometry, uniform stability—are experimentally validated, underscoring parallels to insect circuits. Finally, Webb described honeybee navigation, featuring path integration, vector memories, route optimization, and the famous waggle dance. She proposed that allocentric velocity signals and vector manipulation within the central complex can encode and transmit distances and directions, enabling both sophisticated foraging and inter-bee communication via dance-based cues.

Trackoscope: A low-cost, open, autonomous tracking microscope for long-term observations of microscale organisms

Cells and microorganisms are motile, yet the stationary nature of conventional microscopes impedes comprehensive, long-term behavioral and biomechanical analysis. The limitations are twofold: a narrow focus permits high-resolution imaging but sacrifices the broader context of organism behavior, while a wider focus compromises microscopic detail. This trade-off is especially problematic when investigating rapidly motile ciliates, which often have to be confined to small volumes between coverslips affecting their natural behavior. To address this challenge, we introduce Trackoscope, an 2-axis autonomous tracking microscope designed to follow swimming organisms ranging from 10μm to 2mm across a 325 square centimeter area for extended durations—ranging from hours to days—at high resolution. Utilizing Trackoscope, we captured a diverse array of behaviors, from the air-water swimming locomotion of Amoeba to bacterial hunting dynamics in Actinosphaerium, walking gait in Tardigrada, and binary fission in motile Blepharisma. Trackoscope is a cost-effective solution well-suited for diverse settings, from high school labs to resource-constrained research environments. Its capability to capture diverse behaviors in larger, more realistic ecosystems extends our understanding of the physics of living systems. The low-cost, open architecture democratizes scientific discovery, offering a dynamic window into the lives of previously inaccessible small aquatic organisms.

Hunger state-dependent modulation of decision-making in larval Drosophila

It is critical for all animals to make appropriate, but also flexible, foraging decisions, especially when facing starvation. Sensing olfactory information is essential to evaluate food quality before ingestion. Previously, we found that <i>Drosophila</i> larvae switch their response to certain odors from aversion to attraction when food deprived. The neural mechanism underlying this switch in behavior involves serotonergic modulation and reconfiguration of odor processing in the early olfactory sensory system. We now investigate if a change in hunger state also influences other behavioral decisions. Since it had been shown that fly larvae can perform cannibalism, we investigate the effect of food deprivation on feeding on dead conspecifics. We find that fed fly larvae rarely use dead conspecifics as a food source. However, food deprivation largely enhances this behavior. We will now also investigate the underlying neural mechanisms that mediate this enhancement and compare it to the already described mechanism for a switch in olfactory choice behavior. Generally, this flexibility in foraging behavior enables the larva to explore a broader range of stimuli and to expand their feeding choices to overcome starvation.

Worms use their brain to regulate their behavior and physiology to deal with the lethal threat of hydrogen peroxide

In this talk I will discuss our recent findings that sensory signals from the brain adjust the physiology and behavior of the nematode C. elegans, enabling this animal to deal with the lethal threat of hydrogen peroxide. Hydrogen peroxide (H2O2) is the most common chemical threat in the microbial battlefield. Prevention and repair of the damage that hydrogen peroxide inflicts on macromolecules are critical for health and survival. In the first part of the talk, I will discuss our findings that C. elegans represses their own H2O2 defenses in response to sensory perception of Escherichia coli, the nematode’s food source, because E. coli can deplete H2O2 from the local environment and thereby protect the nematodes. Thus, the E. coli self-defense mechanisms create a public good, an environment safe from the threat of H2O2, that benefits C. elegans. In the second part of the talk, I will discuss how the modulation of C. elegans’ sensory perception by the interplay of hydrogen peroxide and bacteria adjusts the nematode’s behavior to improve the nematode’s chances of finding a niche that provides both food and protection from hydrogen peroxide.

Adapt or Die: Transgenerational Inheritance of Pathogen Avoidance (or, How getting food poisoning might save your species)

Caenorhabditis elegans must distinguish pathogens from nutritious food sources among the many bacteria to which it is exposed in its environment1. Here we show that a single exposure to purified small RNAs isolated from pathogenic Pseudomonas aeruginosa (PA14) is sufficient to induce pathogen avoidance in the treated worms and in four subsequent generations of progeny. The RNA interference (RNAi) and PIWI-interacting RNA (piRNA) pathways, the germline and the ASI neuron are all required for avoidance behaviour induced by bacterial small RNAs, and for the transgenerational inheritance of this behaviour. A single P. aeruginosa non-coding RNA, P11, is both necessary and sufficient to convey learned avoidance of PA14, and its C. elegans target, maco-1, is required for avoidance. Our results suggest that this non-coding-RNA-dependent mechanism evolved to survey the microbial environment of the worm, use this information to make appropriate behavioural decisions and pass this information on to its progeny.

“Wasn’t there food around here?”: An Agent-based Model for Local Search in Drosophila

The ability to keep track of one’s location in space is a critical behavior for animals navigating to and from a salient location, and its computational basis is now beginning to be unraveled. Here, we tracked flies in a ring-shaped channel as they executed bouts of search triggered by optogenetic activation of sugar receptors. Unlike experiments in open field arenas, which produce highly tortuous search trajectories, our geometrically constrained paradigm enabled us to monitor flies’ decisions to move toward or away from the fictive food. Our results suggest that flies use path integration to remember the location of a food site even after it has disappeared, and flies can remember the location of a former food site even after walking around the arena one or more times. To determine the behavioral algorithms underlying Drosophila search, we developed multiple state transition models and found that flies likely accomplish path integration by combining odometry and compass navigation to keep track of their position relative to the fictive food. Our results indicate that whereas flies re-zero their path integrator at food when only one feeding site is present, they adjust their path integrator to a central location between sites when experiencing food at two or more locations. Together, this work provides a simple experimental paradigm and theoretical framework to advance investigations of the neural basis of path integration.

Internal structure of honey bee swarms for mechanical stability and division of labor

The western honey bee (Apis mellifera) is a domesticated pollinator famous for living in highly social colonies. In the spring, thousands of worker bees and a queen fly from their hive in search of a new home. They self-assemble into a swarm that hangs from a tree branch for several days. We reconstruct the non-isotropic arrangement of worker bees inside swarms made up of 3000 - 8000 bees using x-ray computed tomography. Some bees are stationary and hang from the attachment board or link their bodies into hanging chains to support the swarm structure. The remaining bees use the chains as pathways to walk around the swarm, potentially to feed the queen or communicate with one another. The top layers of bees bear more weight per bee than the remainder of the swarm, suggesting that bees are optimizing for additional factors besides weight distribution. Despite not having a clear leader, honey bees are able to organize into a swarm that protects the queen and remains stable until scout bees locate a new hive.

Natural switches in sensory attention rapidly modulate hippocampal spatial codes

During natural behavior animals dynamically switch between different behaviors, yet little is known about how the brain performs behavioral-switches. Navigation is a complex dynamic behavior that enables testing these kind of behavioral switches: It requires the animal to know its own allocentric (world-centered) location within the environment, while also paying attention to incoming sudden events such as obstacles or other conspecifics – and therefore the animal may need to rapidly switch from representing its own allocentric position to egocentrically representing ‘things out-there’. Here we used an ethological task where two bats flew together in a very large environment (130 meters), and had to switch between two behaviors: (i) navigation, and (ii) obstacle-avoidance during ‘cross-over’ events with the other bat. Bats increased their echolocation click-rate before a cross-over, indicating spatial attention to the other bat. Hippocampal CA1 neurons represented the bat’s own position when flying alone (allocentric place-coding); surprisingly, when meeting the other bat, neurons switched very rapidly to jointly representing the inter-bat distance × position (egocentric × allocentric coding). This switching to a neuronal representation of the other bat was correlated on a trial-by-trial basis with the attention signal, as indexed by the bat’s echolocation calls – suggesting that sensory attention is controlling these major switches in neural coding. Interestingly, we found that in place-cells, the different place-fields of the same neuron could exhibit very different tuning to inter-bat distance – creating a non-separable coding of allocentric position × egocentric distance. Together, our results suggest that attentional switches during navigation – which in bats can be measured directly based on their echolocation signals – elicit rapid dynamics of hippocampal spatial coding. More broadly, this study demonstrates that during natural behavior, when animals often switch between different behaviors, neural circuits can rapidly and flexibly switch their core computations.

Three levels of variability in the collective behavior of locusts

Many aspects of collective behavior depend on interactions between conspecifics. This is especially true for the collective motion of locusts, which swarm in millions while maintaining synchrony among individuals. However, whether locusts share and maintain the same socio-behavioral patterns – between groups, individuals and situations – remains an open question. Studying marching locusts under lab conditions, we found that (1) different groups behave differently; (2) locusts within a group homogenize their behavior; and (3) individuals have different socio-behavioral tendencies and context-dependent states. These variability levels suggest that behavioral differences within and among individuals exist, affect others, and shape the collective behavior of the entire group.

The Evolution of Looking and Seeing: New Insights from Colorful Jumping Spiders

During communication, alignment between signals and sensors can be critical. Signals are often best perceived from specific angles, and sensory systems can also exhibit strong directional biases. However, we know little about how animals establish and maintain such signaling alignment during communication. To investigate this, we characterized the spatial dynamics of visual courtship signal- ing in the jumping spider Habronattus pyrrithrix. The male performs forward-facing displays involving complex color and movement patterns, with distinct long- and short-range phases. The female views displays with 2 distinct eye types and can only perceive colors and fine patterns of male displays when they are presented in her frontal field of view. Whether and how courtship interactions pro- duce such alignment between male display and female field of view is unknown. We recorded relative positions and orientations of both actors throughout courtship and established the role of each sex in maintaining signaling alignment. Males always oriented their displays toward the female. However, when females were free to move, male displays were consistently aligned with female princi- pal eyes only during short-range courtship. When female position was fixed, signaling alignment consistently occurred during both phases, suggesting that female movement reduces communication efficacy. When female models were experimentally rotated to face away during courtship, males rarely repositioned themselves to re-align their display. However, males were more likely to present cer- tain display elements after females turned to face them. Thus, although signaling alignment is a function of both sexes, males appear to rely on female behavior for effective communication

Australian Bogong moths use a true stellar compass for long-distance navigation at night

Each spring, billions of Bogong moths escape hot conditions in different regions of southeast Australia by migrating over 1000 km to a limited number of cool caves in the Australian Alps, historically used for aestivating over the summer. At the beginning of autumn the same individuals make a return migration to their breeding grounds to reproduce and die. To steer migration Bogong moths sense the Earth’s magnetic field and correlate its directional information with visual cues. In this presentation, we will show that a critically important visual cue is the distribution of starlight within the austral night sky. By tethering spring and autumn migratory moths in a flight simulator, we found that under natural dorsally-projected night skies, and in a nulled magnetic field (disabling the magnetic sense), moths flew in their seasonally appropriate migratory directions, turning in the opposite direction when the night sky was rotated 180°. Visual interneurons in the moth’s optic lobe and central brain responded vigorously to identical sky rotations. Migrating Bogong moths thus use the starry night sky as a true compass to distinguish geographic cardinal directions, the first invertebrate known to do so. These stellar cues are likely reinforced by the Earth’s magnetic field to create a robust compass mechanism for long-distance nocturnal navigation.

Safety in numbers: how animals use motion of others as threat or safety cues

Our work concerns the general problem of adaptive behaviour in response to predatory threats, and of the neural mechanisms underlying a choice between strategies. When faced with a threat, an animal must decide whether to freeze, reducing its chances of being noticed, or to flee to the safety of a refuge. Animals from fish to primates choose between these two alternatives when confronted by an attacking predator, a choice that largely depends on the context in which the threat occurs. Recent work has made strides identifying the pre-motor circuits, and their inputs, which control freezing behaviour in rodents, but how contextual information is integrated to guide this choice is still far from understood. The social environment is a potent contextual modulator of defensive behaviours of animals in a group. Indeed, anti-predation strategies are believed to be a major driving force for the evolution of sociality. We recently found that fruit flies in response to visual looming stimuli, simulating a large object on collision course, make rapid freeze/flee choices accompanied by lasting changes in the fly’s internal state, reflected in altered cardiac activity. In this talk, I will discuss our work on how flies process contextual cues, focusing on the social environment, to guide their behavioural response to a threat. We have identified a social safety cue, resumption of activity, and visual projection neurons involved in processing this cue. Given the knowledge regarding sensory detection of looming threats and descending neuron involved in the expression of freezing, we are now in a unique position to understand how information about a threat is integrated with cues from the social environment to guide the choice of whether to freeze.

Collective Ecophysiology and Physics of Social Insects

Collective behavior of organisms creates environmental micro-niches that buffer them from environmental fluctuations e.g., temperature, humidity, mechanical perturbations, etc., thus coupling organismal physiology, environmental physics, and population ecology. This talk will focus on a combination of biological experiments, theory, and computation to understand how a collective of bees can integrate physical and behavioral cues to attain a non-equilibrium steady state that allows them to resist and respond to environmental fluctuations of forces and flows. We analyze how bee clusters change their shape and connectivity and gain stability by spread-eagling themselves in response to mechanical perturbations. Similarly, we study how bees in a colony respond to environmental thermal perturbations by deploying a fanning strategy at the entrance that they use to create a forced ventilation stream that allows the bees to collectively maintain a constant hive temperature. When combined with quantitative analysis and computations in both systems, we integrate the sensing of the environmental cues (acceleration, temperature, flow) and convert them to behavioral outputs that allow the swarms to achieve a dynamic homeostasis.

Blood is thicker than water

According to Hamilton’s inclusive fitness hypothesis, kinship is an organizing principle of social behavior. Behavioral evidence supporting this hypothesis includes the ability to recognize kin and the adjustment of behavior based on kin preference with respect to altruism, attachment and care for offspring in insect societies. Despite the fundamental importance of kinship behavior, the underlying neural mechanisms are poorly understood. We repeated behavioral experiments by Hepper on behavioral preference of rats for their kin. Consistent with Hepper’s work, we find a developmental time course for kinship behavior, where rats prefer sibling interactions at young ages and express non-sibling preferences at older ages. In probing the brain areas responsible for this behavior, we find that aspiration lesions of the lateral septum but not control lesions of cingulate cortices eliminate the behavioral preference in young animals for their siblings and in older rats for non-siblings. We then presented awake and anaesthetized rats with odors and calls of age- and status-matched kin (siblings and mothers) and non-kin (non-siblings and non-mothers) conspecifics, while performing in vivo juxta-cellular and whole-cell patch-clamp recordings in the lateral septum. We find multisensory (olfactory and auditory) neuronal responses, whereby neurons typically responded preferentially but not exclusively to individual social stimuli. Non-kin-odor responsive neurons were found dorsally, while kin-odor responsive neurons were located in ventrally in the lateral septum. To our knowledge such an ordered representation of response preferences according to kinship has not been previously observed and we refer this organization as nepotopy. Nepotopy could be instrumental in reading out kinship from preferential but not exclusive responses and in the generation of differential behavior according to kinship. Thus, our results are consistent with a role of the lateral septum in organizing mammalian kinship behavior.

Who can turn faster? Comparison of the head direction circuit of two species

Ants, bees and other insects have the ability to return to their nest or hive using a navigation strategy known as path integration. Similarly, fruit flies employ path integration to return to a previously visited food source. An important component of path integration is the ability of the insect to keep track of its heading relative to salient visual cues. A highly conserved brain region known as the central complex has been identified as being of key importance for the computations required for an insect to keep track of its heading. However, the similarities or differences of the underlying heading tracking circuit between species are not well understood. We sought to address this shortcoming by using reverse engineering techniques to derive the effective underlying neural circuits of two evolutionary distant species, the fruit fly and the locust. Our analysis revealed that regardless of the anatomical differences between the two species the essential circuit structure has not changed. Both effective neural circuits have the structural topology of a ring attractor with an eight-fold radial symmetry (Fig. 1). However, despite the strong similarities between the two ring attractors, there remain differences. Using computational modelling we found that two apparently small anatomical differences have significant functional effect on the ability of the two circuits to track fast rotational movements and to maintain a stable heading signal. In particular, the fruit fly circuit responds faster to abrupt heading changes of the animal while the locust circuit maintains a heading signal that is more robust to inhomogeneities in cell membrane properties and synaptic weights. We suggest that the effects of these differences are consistent with the behavioural ecology of the two species. On the one hand, the faster response of the ring attractor circuit in the fruit fly accommodates the fast body saccades that fruit flies are known to perform. On the other hand, the locust is a migratory species, so its behaviour demands maintenance of a defined heading for a long period of time. Our results highlight that even seemingly small differences in the distribution of dendritic fibres can have a significant effect on the dynamics of the effective ring attractor circuit with consequences for the behavioural capabilities of each species. These differences, emerging from morphologically distinct single neurons highlight the importance of a comparative approach to neuroscience.

Untangling the web of behaviours used to produce spider orb webs

Many innate behaviours are the result of multiple sensorimotor programs that are dynamically coordinated to produce higher-order behaviours such as courtship or architecture construction. Extendend phenotypes such as architecture are especially useful for ethological study because the structure itself is a physical record of behavioural intent. A particularly elegant and easily quantifiable structure is the spider orb-web. The geometric symmetry and regularity of these webs have long generated interest in their behavioural origin. However, quantitative analyses of this behaviour have been sparse due to the difficulty of recording web-making in real-time. To address this, we have developed a novel assay enabling real-time, high-resolution tracking of limb movements and web structure produced by the hackled orb-weaver Uloborus diversus. With its small brain size of approximately 100,000 neurons, the spider U. diversus offers a tractable model organism for the study of complex behaviours. Using deep learning frameworks for limb tracking, and unsupervised behavioural clustering methods, we have developed an atlas of stereotyped movement motifs and are investigating the behavioural state transitions of which the geometry of the web is an emergent property. In addition to tracking limb movements, we have developed algorithms to track the web’s dynamic graph structure. We aim to model the relationship between the spider’s sensory experience on the web and its motor decisions, thereby identifying the sensory and internal states contributing to this sensorimotor transformation. Parallel efforts in our group are establishing 2-photon in vivo calcium imaging protocols in this spider, eventually facilitating a search for neural correlates underlying the internal and sensory state variables identified by our behavioural models. In addition, we have assembled a genome, and are developing genetic perturbation methods to investigate the genetic underpinnings of orb-weaving behaviour. Together, we aim to understand how complex innate behaviours are coordinated by underlying neuronal and genetic mechanisms.
